Lab 02
Today, we learned about 1) Smear (liquid and solid) 2) Simple Stain and 3) Gram Stain
All the different stains start with a smear – the procedure is simple – take whatever bacteria, put on slide, wait till it’s dry, then heat fix it. Simple Stain is only using one single dye (we used Crystal Violet).
Gram Stain is more complex, and it involves 1) Crystal violet for 20 seconds then rinse 2) Gram Iodine for 1 minute then rinse 3) EtOH, then rinse 4) Counter stain with Safirin, then rinse.
All my 6 slides turned out okay — the only one that’s Gram Negative is … Pseudomonas aeruginosa. Bascillus cereus, Staphylococcus aureus, Mycobacterium smegmatis are all gram positive.
